Role of dprA in transformation of Campylobacter jejuni.

چکیده
The role of a dprA ortholog (Cj0634) in Campylobacter jejuni transformation was phenotypically assessed using two strains. C. jejuni strain 11168 was naturally competent for transformation by chromosomal DNA, while efficiency decreased 100-fold in a Cj0634::aphA mutant, whereas C. jejuni strain 480 was not naturally competent. C. jejuni strain 480 but not 11168 could be electro-transformed by shuttle plasmid pRY111, an effect completely abolished by Cj0634 interruption. Complementation of the Cj0634 mutation in C. jejuni strain 480 in trans with vectors containing the dprA homologs from C. jejuni, Helicobacter pylori, or Haemophilus influenzae, completely (for Cj0634) or partially (H. pylori>H. influenzae) restored electro-transformation. Thus, C. jejuni expresses a DprA ortholog that functionally most closely resembles that of H. pylori and is involved in DNA transformation.

The Role of Campylobacter Jejuni in Campylobacteriosis
Campylobacter jejuni is a Gram negative, curved, motile, thermophilic and microaerophilic bacillus in Campylobacteriaceae family, which is one of the main causes of enteritis in human named campylobacteriosis. The main reservoir of Campylobacter jejuni is the alimentary tract of animals, especially chicken and turkey. Consumption of semi-cooked chicken meat, raw milk and unchlorinated water are...
متن کاملRapid Detection of Campylobacter jejuni by Polymerase Chain Reaction and Evaluation of its Sensitivity and Specificity
Introduction: Campylobacter jejuni is one of the most common causes of food poising in humans. Rapid and specific detection of these bacteria has an important role in diagnosis and treatment of infection. The aim of this study was to design a specific PCR for the detection of Campylobacter jejuni. Methods: In this experimental study, oxidoreductase gene from the Campylobacter jejuni was sele...
متن کاملAssess the prevalence rate of Campylobacter genus and Campylobacter jejuni species in raw milk collected from the Amol City by Multiplex- Polymerase Chain Reaction
Background & Objective: Campylobacter can be transmitted through the raw milk. The purpose of this study was to determine the prevalence of Campylobacter genus and Campylobacter jejuni (C. jejuni) species in raw milk samples. Materials & Methods: In this study, 72 samples of raw milk were collected of the platforms milk in the Amol city in summer. Phenotypic identification of Campylobacter genu...
متن کاملDetection of cytolethal distending toxin (cdt) Genes in Campylobacter jejuni and Campylobacter coli isolated from the intestinal of commercial broiler chickens, turkey and quail of Iran
Background: Campylobacter jejuni and Campylobacter coli are zoonotic bacteria which are frequently associated with human diarrhea. Sharing of the cytolethal distending toxin (cdt) genes in Campylobacter is common and is considered species specific. OBJECTIVES: In this study we focused on detecting the presence of cdt gene in C. jejuni and C. coli isolated from broilers, turkeys and quails of Ir...
متن کاملThe prevalence of campylobacter jejuni induced gastroenteritis in patients with diarrhea referring to Mean Reza hospital in Mashhad,Iran
The ever- increasing importance of campylobacter jejuni infectious is due to its associations with severe diarrhoea , especially in children. In order to determine the bacterial etiology of gastroenteritis, we cultured the stool of 903 patients over 10 months from April 1993 to February 1994. In this study, campylobacter jejuni, salmonella and Shigella were isolated from 19,29 and 16 patient...
